Human ADPKD and Normal Human Kidney Biomaterials and Primary Cells

HUMAN ADPKD AND NORMAL HUMAN KIDNEY BIOMATERIALS AND PRIMARY CELLS AVAILABLE FOR RESEARCH:

ADPKD kidneys are obtained at the time of nephrectomy (removal of the kidney pre- or post-transplantation). Therefore, the kidneys are retrieved from individuals with stage 4-5 chronic kidney disease (CKD). In some instances, normal human kidney tissues are obtained from non-transplanted kidneys harvested by a transplant network and perfused with a preservation solution but judged to be unsuitable for transplantation due to donor factors, such as anatomical problems, biopsy findings, and cold ischemic time. The Cores consider several factors, such as KDPI (an indicator of relative risk score for graft survival) before accepting cadaver kidneys for research. In other instances, kidney tissues are obtained from the non-affected regions during a radical or partial nephrectomy to treat kidney cancer. This source of normal kidney tissues depends on the specific Core of three Research and Translational Core Centers (RTCCs). The Cores may offer frozen and formalin (or paraformaldehyde)-fixed paraffin-embedded tissues, and primary cultures of epithelial cells from ADPKD kidneys and/or normal kidneys to assist investigators in translational research. Biospecimens, such as cyst fluid, serum, or urine, may also be available for some participants.

AVAILABLE BIOMATERIALS:
· Frozen ADPKD tissues (single or multiple cysts, snap frozen in LN2)
· Frozen normal human kidney (NHK) tissues (cortex or medulla, snap frozen in LN2)
· ADPKD tissue (formalin or para-formaldehyde-fixed paraffin-embedded) sections on glass microscope slides for immunohistochemistry
· NHK tissue (formalin or para-formaldehyde-fixed paraffin-embedded) sections on glass microscope slides for immunohistochemistry
· ADPKD or NHK tissue microscope slides stained with hematoxylin and eosin or Masson Trichrome
· ADPKD cyst fluid from individual cysts or pooled fluid from multiple cysts, depending on the RTCC
· Primary cultures of ADPKD cells from multiple surface cysts or individual cysts, depending on the RTCC
· NHK cells cultured from renal cortex to outer medulla
· NHK cells cultured from outer medulla to initial inner medulla
· In some cases, primary ADPKD cells, cyst fluid, frozen and fixed tissue available from single unique cysts
· In some cases, blood and urine were collected at the time ADPKD tissues were obtained by nephrectomy
